Abstract 


 In this study, we developed a technique to identify rapid sequential activation patterns in the medial prefrontal cortex (mPFC) using one-photon calcium imaging during a spatial memory task. We examined the relationship between these patterns and animal behavior, as well as their stability over several days of learning.

 Mice performed an olfaction-guided spatial memory task in a three-arm arena, associating odors in the middle arm with rewards in the left or right arms. We used hierarchical clustering to group sequences, observing an increase in sequence rates and cluster numbers during learning. Diversity indices suggested the emergence of a dominant sequence pattern post-learning.

 Our findings demonstrate that one-photon recordings enable the analysis of fast mPFC sequences, which vary in rate and content based on task phase. This suggests a potential role for mPFC sequential activation in working memory tasks.
